HIV-infection

Human immunodeficiency virus (HIV) is a lentivirus that causes acquired immunodeficiency syndrome (AIDS), a condition in humans, in which progressive failure of the immune system allows life-threatening opportunistic infections and cancers to thrive. Infection with HIV occurs by the transfer of blood, semen, vaginal fluid, pre-ejaculate or breast milk. Within these bodily fluids, HIV is present as both free virus particles and virus within infected immune cells.

There are two types of HIV - HIV-1 and HIV-2. In the United States, unless otherwise noted, the term “HIV” primarily refers to HIV-1. Both types of HIV damage a person’s body by destroying specific blood helper T cells (CD4+), which are crucial to helping the body fight diseases.

HIV infects also other vital cells in the human immune system such as macrophages and dendritic cells. HIV infection leads to low levels of CD4+ T cells through three main mecha-nisms: first - direct viral killing of infected cells; second - increased rates of apoptosis in infected cells and third - killing of infected CD4+ T cells by CD8 cytotoxic lymphocytes that recognize infected cells. When CD4+ T cell numbers decline below a critical level, cell-mediated immunity is lost and the body becomes progressively more susceptible to opportunistic infections. Most untreated people infected with HIV-1 develop AIDS. These individuals mostly die from opportunistic infections or malignancies associated with the progressive failure of the immune system.

DNA-HIV PCR kit (TR-V0-G) is qualitative DNA test based on the amplification of HIV DNA target region and endogenous control - human fragment of β-globin gene. Such Internal Control allows to determine sufficient quantity equivalent to that of cells in the clinical sample. 

HIV-Monitor PCR kit (R-V0-MC) is RNA based test for qualitative detection and quantitation of HIV type 1 RNA in the clinical material (plasma). The kit contains Internal Control, based on the β-globin gene.

Analytical sensitivity is 100 GE/ml DNA HIV-1 per 250 μl of clinical sample.

The linear range of HIV-Monitor PCR kit is 500 - 5.000.000 copies per 1 ml (if the sample volume is 100 μl) or 50 - 500.000 copies per 1 ml (if the sample volume is 1 ml).

Kits contain DNA/RNA isolation system (DNA-HIV), reverse transcription (HIV-Monitor version) and DNA/cDNA amplification systems.

For detection, 2 channels - FAM/Green and JOE/Yellow/HEX are needed.
